四六級英語考試口語考試的的每場考試都由兩名主考官主持、三名考生參加，時間為20分鍾。考試由三部分組成：
第一部分主要是「熱身練習」，先由考生作自我介紹，目的是使考生進入一個良好的應考狀態，以充分發揮出自己的英語口語水平；然後主考官向每一個考生提問，所提問題由口語考試所決定的話題而決定。每位考生回答一個問題。時間約5分。
第二部分是考試的重點部分，主要考核考生用英語進行連貫的英語口語口頭表達能力，以及傳達信息、發表意見、參與討論和進行辯論等口頭交際能力。主考官先向每位考生提供文字或圖片等形式的提示信息，考生可准備一分鍾。然後要求考生就所得材料與信息做90秒的發言。考官要求考生根據其發言內容，就規定話題進行小組討論，盡量取得一致結論，時間共約10分。
第三部分由考官再次提問以進一步確定考生的口頭交際能力。時間為5分。
根據四六級考試改革進程，大學英語四、六級考試口語考試仍將與筆試分開實施，繼續採用已經實施了五年的面實行的四、六級口語考試（CET-SET）。同時，考委會將考慮計算機化口語測試，推動大學英語口語教學 。
國家教育部高等教育司為檢驗與認定高校非英語專業在校生對英語口語掌握的程度，全國大學英語四六級考試委員會在1999年11月開始施行的大學英語四六級口語測試。全稱叫大學英語四、六級口語考試（CET Spoken English Test）簡稱大學英語口語考試（CET-SET）。
考試時間
改革後的全國大學英語四、六級考試，四級成績為550分以上（含550 分）或六級成績為520分以上（含520分）。
2014年11月進行的全國大學英語四、六級口語考試的報考條件發生了變化：2005年6月及以後的紙筆或網考四級成績為500分以上（含500分），或六級成績為425分以上（含425分）即可報考。
